Cortisol stimulates gluconeogenesis by increasing the proteolysis (breakdown of proteins) of muscle tissue. Cortisol effect destabilizes the insulin metabolism causing a decrease in tissue sensitivity to this hormone and disturbs the proper functioning of the thyroid gland by lowering the metabolic rate. Elevated levels of cortisol in the blood cause a specific displacement of fat tissue deposits on neck, face and abdominal area. In addition, cortisol has a negative effect on the duration and quality of sleep, thus disturbing normal regeneration.

The herb contains many active ingredients, including flavonoids and withanolides. Forskolin is a substance that, by the activation of the adenylyl cyclase enzyme, increases the level of a cyclic AMP (cAMP). The cAMP is an important secondary messenger in a normal biological response of cells to hormones and other extracellular signals or in communication within the hypothalamic/pituitary axis and feedback control through the induction of factor gene transcription of corticotropin-releasing factor.

Researchers found that obese people have a lower production of cyclic AMP than normal. For these reasons, forskolin can be theoretically speaking a weight-reducing agent, especially among people with reduced cyclic AMP production. Studies in rats have shown that forskolin supplementation does not cause clinically significant adverse effects and can limit weight gain in overweight animals as well as promote weight reduction. In addition, the results of in vito studies show that forskolin stimulates fat metabolism, which in combination with an energy-deficit diet may effectively facilitate reduction of body fat.

7-Keto is a steroid produced in the metabolism process of dehydroepiandrosterone (DHEA). 7-Keto-DHEA is not immediately converted to testosterone or estrogen and is therefore probably more useful than DHEA, which can negatively affect the hormonal balance.

7-Keto reduces the activity of 11β-hydroxysteroid dehydrogenase (11betaHSD) – an enzyme that converts inactive cortisone to active cortisol. 11BetaHSD participates in the metabolism of 7-Keto, as a result of which, the amount of the enzyme capable of converting the cortisone into an undesirable cortisol is limited.

7-Keto has thermogenic properties due to the activation of the following enzymes: acyl-CoA oxidase, glycerol-3-phosphate dehydrogenase, and malate enzyme. Activation of these biocatalysts directs the energy substrates towards the less efficient ATP production in relation to a heat production and promotes the use of fat deposits for energy production. The publications available also state that 7-keto-DHEA supplementation in overweight people combined with a limited- calorie diet, effectively reverses the decline in resting metabolic rate (RMR).
